SYRE Rallies 44.9% in 3 Months. Can Clinical Catalysts Sustain It?

Read MoreHide Full Article

Key Takeaways

  • Spyre gained 44.9% in three months as positive UC data and 2026 readouts drew interest.
  • SPY002 posted 33% clinical remission and 42% endoscopic improvement in phase II ulcerative colitis data.
  • A dense 2026 data calendar faces added pressure from competition and a valuation of 9.45X book value.

Spyre Therapeutics (SYRE - Free Report) has advanced 44.9% in the past three months, far outpacing the Zacks sub-industry’s 11.7% gain and the S&P 500’s 1.2% rise. That run has put the durability of the stock’s momentum in focus.

Positive ulcerative colitis data and several 2026 readouts support investor interest. At the same time, a clinical-stage pipeline and elevated valuation leave less room for disappointing efficacy, safety or differentiation.

Spyre's 3-Month Rally Meets a Catalyst-Heavy Setup

SPY002 delivered positive 12-week induction results in Part A of the phase II SKYLINE study. The anti-TL1A antibody produced a statistically significant 10.7-point reduction in Robart’s Histopathology Index, a measure of tissue-level inflammation in ulcerative colitis.

The study also showed a 33% clinical remission rate and 42% endoscopic improvement rate. SPY002 was generally well tolerated, with no drug-related serious adverse events reported, giving investors clinical evidence to weigh alongside the recent share-price move.

SYRE Has More Phase II Readouts Ahead in 2026

The next catalyst is Part A top-line data for SPY003 in ulcerative colitis, expected in the third quarter of 2026. SKYLINE Part B is also enrolling patients across monotherapy and combination arms, with induction data expected in 2027.

Beyond inflammatory bowel disease, the phase II SKYWAY study has completed enrollment across rheumatoid arthritis, psoriatic arthritis and axial spondyloarthritis. Rheumatoid arthritis data are due in the third quarter, followed by psoriatic arthritis and axial spondyloarthritis results in the fourth quarter.

Spyre's Early UC Data Raise the Bar for Expectations

SPY001 and SPY002 have both generated encouraging phase II ulcerative colitis signals, while the broader SKYLINE strategy tests three validated inflammatory targets as monotherapies and pairwise combinations. Continued progress could strengthen the case for deeper, less frequent treatment approaches.

Competition also raises the differentiation threshold. AbbVie (ABBV - Free Report) continues to generate IBD data for Skyrizi and Rinvoq. Johnson & Johnson (JNJ - Free Report) is advancing JNJ-4804, a co-antibody therapy targeting IL-23 and TNF, after phase IIb results in ulcerative colitis and Crohn’s disease.

SYRE's Valuation Leaves Less Room for Clinical Missteps

SYRE trades at 9.45X trailing 12-month book value per share versus a five-year median of 2.71X. The premium puts more weight on upcoming readouts because the company still has no approved products or commercial revenues.

Disappointing efficacy, unexpected safety issues or insufficient differentiation could pressure the shares. Intense competition adds another layer of risk, even as Spyre’s multiple programs reduce dependence on any single experimental therapy.

SYRE's Mixed Scores Temper the Momentum Signal

SYRE’s setup remains balanced. A dense clinical calendar can keep attention on the pipeline, but valuation and development risk make execution across the next several readouts important.

The stock currently carries a Zacks Rank #3 (Hold). Its Momentum Score of A is favorable, but the Value Score of F, Growth Score of F and VGM Score of F show that the stock does not have broad support across the Zacks Style Scores. The combination tempers the momentum signal without establishing a clear directional call. You can see the complete list of today’s Zacks #1 (Strong Buy) Rank stocks here.
