TNGX Stock Falls 16% Despite Positive Pancreatic Cancer Study Data

Read MoreHide Full Article

Key Takeaways

  • Tango Therapeutics shares fell 16.4% as pancreatic cancer ORR declined to 82% from June's 92% readout.
  • The combination delivered 88% nine-month PFS and 100% disease control among 22 evaluable patients with PDAC.
  • Tango plans a phase III pancreatic cancer study in 2027 and will cut its workforce by approximately 14%.

Tango Therapeutics (TNGX - Free Report) shares fell 16.4% on Friday after updated phase I/II study data for vopimetostat plus daraxonrasib in previously treated pancreatic cancer showed an 82% objective response rate (ORR), down from 92% in June's initial readout. The sell-off appeared to reflect investor disappointment with the lower response rate, although the latest analysis showed durable disease control, including an 88% progression-free survival (PFS) rate at nine months. The results were presented at the 2026 European Society for Medical Oncology Congress.

Strategic changes may also have weighed on sentiment. Per Tango’s latest regulatory filing, the board approved a research and development reprioritization last week. The plan prioritizes PRMT5 programs vopimetostat and TNG456, deprioritizes novel-target discovery and includes a workforce reduction of approximately 14%, mostly in research, plus discontinuation of certain preclinical and research activities.

Tango’s vopimetostat is an investigational, once-daily oral PRMT5 inhibitor. The phase I/II dose-escalation and expansion study is evaluating the candidate with Revolution Medicines’ RAS(ON) inhibitors, daraxonrasib or zoldonrasib, in patients with MTAP-deleted, RAS-mutant pancreatic ductal adenocarcinoma (PDAC) or non-small cell lung cancer (NSCLC). The latest analysis had an August 20, 2026, data cutoff.

Updated Data Readout Shows Durable Response in Pancreatic Cancer

Tango reported that among 22 evaluable patients receiving vopimetostat plus daraxonrasib as second- or third-line treatment for PDAC, 18 responded, resulting in an ORR of 82%. This included 11 confirmed partial responses, while the remaining seven responses, initially unconfirmed at the data cutoff, were confirmed subsequently. The disease control rate was 100%. In June, Tango reported a 92% ORR among 12 patients, so the rate was lower in the expanded dataset despite the larger sample.

Median PFS had not been reached, and the nine-month PFS rate was 88% compared with a six-month PFS rate of 90% in June. Median follow-up in the latest analysis was 7.1 months, and 13 of the 22 evaluable patients received the combination as third-line treatment. The results suggest potentially durable responses but remain early-stage findings from a small group.

Year to date, TNGX stock has skyrocketed 163.4% compared with the industry’s 4.7% growth.

At the 200-mg vopimetostat dose level, the combination was generally tolerated among 25 safety-evaluable patients. There were no dose-limiting toxicities and no withdrawals due to treatment-related adverse events, which were mostly mild to moderate in severity. However, the 250-mg dose level was not well tolerated, with three dose-limiting toxicities in two of four safety-evaluable patients.

Tango also reported vopimetostat plus zoldonrasib results in MTAP-deleted, KRAS G12D-mutant PDAC. Among 35 evaluable patients, the combination showed a 54% ORR, a 61% nine-month PFS rate and a 97% disease control rate. Across 39 safety-evaluable patients, there were no dose-limiting toxicities or withdrawals due to treatment-related adverse events.

TNGX’s Next Steps

Tango plans to initiate a global, randomized registrational phase III study of vopimetostat plus daraxonrasib in first-line MTAP-deleted PDAC in the first half of 2027. The study will compare once-daily vopimetostat 200 mg plus daraxonrasib 100 mg with standard-of-care chemotherapy. The dual primary endpoints of the planned late-stage study are ORR and PFS, with overall survival as a key secondary endpoint.

The company also plans to discuss a potential second-line PDAC regulatory pathway with authorities and is pursuing ways to secure daraxonrasib supply for late-stage development. A phase I/II study of vopimetostat plus a RAS inhibitor in second-line, MTAP-wild-type PDAC is also planned for the first half of 2027.

TNGX’s Vopimetostat Also Shows Early Activity in NSCLC

Tango reported that among five evaluable NSCLC patients receiving vopimetostat plus daraxonrasib at dose level 1, four had partial responses, resulting in an 80% ORR with a disease control rate of 100%. The small sample limits conclusions and requires confirmation in further studies.

No serious treatment-related adverse events were reported in the NSCLC group. Treatment-related adverse events were mostly mild to moderate in severity. Tango expects to report separate phase I/II vopimetostat monotherapy data in NSCLC in January 2027.
